Procedure Overview
 
REAGEN L-CarnitineTest Kit assay transfers an acetyl group from CoA to Carnitine and the free CoA formed is further processed with substrate to give absorbance (**5 nm).

Required Materials Not Provided With the Kit
 
  • Microtiter plate reader (4*5 nm)
  • Vortex mixer
  • *-*0ul,  **-2*0ul and***-***0uL pipettes
  • Perchloric acid(AR)
  • KOH(AR)
  • pH meter
  • filter paper 
 
 
Reagent preparation
 
  • Perchloric acid(*3%): *3ml perchloric acid add *7ml water,mix.
  • KOH(4mol/L):*2.4g KOH add water to **0ml.
  • Carnitine Substrate(1x):get 0.5ml 5xCarnitine Substrate,add 2ml water,mix.
  • Enzyme(CAT):get *0ul to centrifuge,Remove the supernatant,add **0ul water,mix.
  • Prepare the standards:add 0.5ml deionized  water to  Carnitine Standard bottle.Dilute*4ug/ml to*6,9.6,6.4,3.2,1.6 ug/ml

Sample Preparation
Milk powder
1) 2.5 gsample,add*5 mL *0℃water ,mix.。
2) Add5 mL *3% Perchloric acid,mix,stay *0mins.Add water to50ml,mix,filter to get the filtrate(must be clear solution)。
3) Get*0 mL filtrate,use 4 mol/L KOH to adjust pH *2.***3,*0ËšC water bath*0 min。Cool to room temperature,use *3% Perchloric acid to adjust pH to 7.0~7.5,
4) Add water to*0ml.mix,let stay for*2h in 4℃,getsupernatant to test.
Dilution factor: *0. (the filtrate must be clear)
 
 
Test protocol
 
1) Setfivestandard wells(1.6,3.2,6.4,9.6,*6)、sample well,
2) Add*00ul standard or sample to the Micro-well,
3) Add*0ul substrate,
4) Add *0ul water,At room temperature ,After 5 mins,**5nm detect OD1.
5) Add *0ul CAT,At room temperature ,After*0 mins,**5nm detect OD2
 
Concentration Calculations
A standard curve can be constructed by plotting the mean relative absorbance (%) obtained from each reference standard against its concentration in ng/mL on a  curve.
OD2-OD1 as the Vertical ordinate,concetration as the Horizontal coordinate.
