1. INTENDED USE
Quicking Melamine Rapid Test is a competitive immunoassay for the semi-quantitative detection of the presence of melamine residue in milk, milk powder, feed and egg.
Detection Limit:
Milk, 1.0 ppm; Milk Powder, 2.5 ppm; Feed, 2.5 ppm; Egg, 1.0 ppm.
Assay Time: 5 - *0 min.
2. PRINCIPLE OF THE ASSAY
Quicking Melamine Rapid Test is based on competitive lateral flow immunochromatographic assay. The Melamine conjugate in the test zone will capture the immuno-gold (colloid gold-Melamine antibody conjugate), when there is very little dissociative Melamine in the samples. A visible red test band indicates a negative result when the control line (C zone) shows that the card is valid. The test band (T zone) will be not visible if Melamine is present in concentration of the cut-off value and above which explains a positive result.

4. SAMPLE EXTRACTION
4.1 Milk (1.0 ppm)
Transfer 0.2 mL of milk sample into a centrifugal tube. (Tip: The volume of milk sample reaches the scale of the small pipette provided in the foil pouch). Add 0.6 mL of assay buffer into the tube and mix well with the milk sample. Use the mixture in the assay.
4.2 Milk powder (2.5 ppm)
Dissolve 0.5 g of milk powder with 5 mL of *0 ℃ tap water. Keep it cool to room temperature. Use the liquid milk for running the assay.
4.3 Feed (2.5 ppm)
Grind a representative sample to the particle size of fine instant coffee (*0% passes through a *0 mesh screen). Weigh out 0.5 g ground portion of the sample into a sample tube. Add 4 mL of assay buffer into the tube and shake for 5 min. Use the supernatant in the assay. (If it is cloudy, do a centrifugation at ***0 rpm for 1 min.)
4.4 Egg (2.5 ppm)
Suck 0.2 mL of egg sample into a centrifugal tube. Add 0.8 mL of assay buffer into the tube and mix well with the sample. Use the mixture in the assay.
5. TEST PROCEDURE
- Take out the cassette from the foil pouch and place it horizontally.
- Gradually drip 3 drops of sample extraction into the assay sample hole.
- Interpret the result in 5 - *0 min. Result after *0 min is considered as invalid.
6. INTERPRETATION OF RESULTS
Positive: Only one clear band in C zone indicates a positive result. Positive shows that the concentration of Melamine is at or above the cut-off value in the sample.
Negative: The presence of both clear band in C zone and T zone.
Invalid: No colored band appears in C zone.
