TTX ELISA KIT General Description The REAGEN®***** direct competitive ELISA is an immunoassay for the quantitative and sensitive detection of ***** in water samples. A pre-sample concentration is not required. If necessary, positive samples can be confirmed by HPLC, protein phosphatase assay or other conventional methods. ASSAY PRINCIPLES The TTX plate kit is a competitive enzyme-labeled ***** sample extract and calibrators are pipetted into the test wells followed by TTX antibody- HRP conjugate into the test wells to initiate the reaction. After this *0 minute incubation the wells are washed with wash solution. a clear substrate is added to the wells and any bound enzyme conjugate causes the conversion to a blue color. After the ****2 minute incubation, the reaction is stopped and the amount of color in each well is read. The color of the unknown samples is compared to the color of the calibrators and the TTX concentration of the samples is derived. REAGENTS AND MATERIALS PROVIDED The kit in its original packaging can be used until the end of the month indicated on the box label when stored at *- 8ºC. 1. Plate containing *2 test strips of 8 wells each vacuum-packed in aluminized pouch with indicating dessicant. 2. 5 vials each containing 1 mL of TTX calibrators corresponding to 0, 5, *0, *0 and **0 ppb. 3. 1 vial containing *0 uL of TTX antibody- HRP conjugate concentrate **0 X. Needs to be diluted prior to use. 4. 1 vial containing 8mL of TTX antibody- HRP conjugate Diluent 5. 1 vial containing *2 mL of Substrate. 6. 1 vial containing 6 mL of Stop Solution. (Caution! 1N H2SO4. Handle with care.) 7. 1 vial containing *0 mL of Wash Solution concentrate *0X. Needs to be diluted prior to use. 8. Instructions
